Cellulases were separated by polyacrylamide disc gel electrophoresis using 7%polyacrylamide gel.［16，17］The gel was stained with Amido Black and destained in 7%acetic acid.According to the colour of protein bands in stained gel，the unstained control gels were cut into slices which were extracted with distilled water，dialysed against water and lyophilized.［9］To estimate the molecular weights of cellulase components，5%，6%，7%and 8%polyacrylamide gels were used with bovine serum albumin，peroxidase，pepsin，amylase，lactate dehydrogenase and creatine kinase as refer ences.The SDS-gel electrophoresis［2］was also used to determine the molecular weight of C1.Cellulose acetate film electrophoresis，polyacrylamide gel electrophoresis and immunoelectrophoresis were employed to determine the purity of C1 cellulase.［9］Antiserum against cellulase was prepared by injecting rabbits with 2 mg of partly purified cellulase in a Freund's adjuvant（1∶1 v/v）for the first injection，0.75 mg and 1 mg of cellulase for the second and third injections，respectively，at 10 day intervals.(https://www.daowen.com)
