Induction and Regulation of Cellulase Forma Tion in Trichoderma.Ⅳ.Changes in Regu Latory Mechanisms of Cellulase Synthesis of Two Mutants with High Cellulase Yields——A Preliminary Analysis of the Mechanism of Increase in Cellulase Production[1]

Two mutants with high cellulase yields of Trichoderna pseudokiningii Rafai EA3-867 and N2-78,showed small colonies on agar plates with synthetic medium or potato-glucose medium, and grew slowly, but on agar plates with peptone-yeast extract medium the small colonies converted into colonies as large as wild strains. The cellulase production of EA3-867 and N2-78 on either cellulose or glycerolsophorose media was higher than that of their wild parents 1096 and MO3,respectively.The inductive activity for cellulase production of washed mycelia of two mutants was also enhanced. The cellulase components, induced by sophorose,of washed mycelia of the mutants and wild strains showed no significant differences,at least in the patterns of polyacrylamine gel electrophoresis. But the protein bands of cellulase of the mutants were considerably deeper in colour than that of their wild parents. It is evident that the increase in cellulase production of both mutants results from the changes in the regulatory mechanism of cellulase systhesis,i.e.,the mutants showed higher responsiveness to inducer and lower susceptibility to catabolite repression than those of their wild parents.
