Detection of CRM197 and wt Alleles by Allele-Specific PCR
PCR product patterns in allele-specific PCR analysis of the template mixtures，which contain different percentages ofwild-type and CRM197 mutant（CRM-Ref.，C7（β197）tox-double lysogen［13］）DNAs，are presented as shown in Figure 4.The estimated sensitivity of the allele-specific assay，with the PCR conditions used，is about 5%of the particular template DNA in the sample.With lower template amounts，the allele-specific PCR products were barely detectable on the gel.
