1.10.2　细胞的免疫荧光

（1）甩片：（2000rpm×10min）将2×104细胞甩在载玻片上；

（2）固定：使用4%paraformaldehyde固定液固定10min，PBS清洗5min×2次，0.1% triton X-100（10μl triton X-100溶于10ml PBS配制）孵育5min，PBS清洗5min×2，5%；

（3）封闭BSA：封闭1h；(https://www.daowen.com)

（4）孵育一抗：BrdU（mouse，1∶1000，BD，552598，USA）4℃孵育过夜；PBS清洗5min×3次；

（5）孵育二抗：goat anti-mouse IgG antibody coupled with Alexa Fluor 594（Invitrogen，Carlsbad，A11032，CA，1∶1000），避光孵育1h，PBS清 洗5min×3次，滴 加DAPI（4′，6′diamidino-2-phenylindole）staining（Vectashield，Vector Laboratories，Inc H1000）封片。

（6）照相：荧光显微镜观察照相。
