Plasmid,M13 Phage,and Synthetic Oligonucleotide Pr...
The plasmid,M13 phage, and oligonucleotide probes for varous genes coding for LH and RC proteins and for bacteriochlorophyll and carotenoid biosynthetic enzymes were prepared as previously described[39]and are listed as shown in Table 1 and Figure 1.
Table 1 Phage M13,plasmids,and oligonucleotides used for probing photosynthetic genes of R.capsulata
ⓐSingle-stranded probes synthesized from M13 DNA were complementary to known open reading frames based on DNA sequence analysis.

Figure 1 Photosynthetic gene cluster map.BamHI restriction sites are shown within a 46-kb photosynthetic gene cluster from R.capsulatus carried by the R'plasmid pRP404[24,36,41].The genes coding for LH-Ⅰ(β and α),RC-L,and RC-M subunits and an open reading frame C2397,are located in the BamHI C fragment, whereas the gene coding for the RC-H subunit resides in the BamHI F fragment. The arrows indicate the direction of transcription. The BamHI fragments between these two genetic loci contain a number of bch genes(in BamHI-D and-E)and crt genes(in BamHI-H,-G,-M,and-J).The shaded areas indicate the genes determined by cluster point mutations conferring the same phenotype. The blank areas represent genes determined by transposon mutagenesis and complementation
