Detection of PCR Products

The amplified HIV1 RNA gag region delineated by the primer pairs SK38/39 was detected by solution olingonucleotide hybridization.The PCR product（10 μl）was hybridized with32 P-la-belled SK19 probe（（3-5）×105cpm，107com/pmol）at 55℃for 15 min.The hybridized gag sequence was separated by electrophoresis in a native 12%polyacrylamide gel and then exposed to X-ray film with intensifying screens for one hour.The bands corresponding to the HIV1 sequence（115 bp）were either cut and counted in a scintillation counter or quantitated with an“Ambis Radioanalytic Imaging System”（Ambis Systems，San Diego，CA）.(https://www.daowen.com)
