2.4 Mitochondrial genome annotation
Genes in O.sinensis mt genome were predicted by MFannot,RNAweasel(http://megasun.bch.umontreal.ca/RNAweasel/)and BLASTn against NCBI Organelle Genome Resources(https://www.ncbi.nlm.nih.gov/genome/organelle/)(Altschul,1990;Lang et al.,2007;Valach et al.,2014).The protein-coding genes(PCGs)and rRNA genes were identified by MFannot and checked by BLASTn against Hypocreales(Altschul,1990),while the tRNA genes were checked using RNAweasel.Intronexon boundaries of the PCGs were adjusted manually on the basis of BLASTn against multiple Hypocreales mt coding sequence(Altschul,1990).The nucleotide sequences of PCGs were translated to protein sequences using the Mold,Protozoan and Coelenterate Mitochondrial code(transl_table=4).Open reading frames(ORFs>100bp)in the intergenic and intronic regions were predicted by MFannot(Valach et al.,2014).Predicted ORFs were analyzed by InterProScan(http://www.ebi.ac.uk/interpro/).The mitochondrial genome map was generated with Circos software(Krzywinski et al.,2009)and CLC Sequence Viewer 7.8.1,and then modified by Adobe Illustrator CC 2015(Version 19.0.0.44,Adobe,San Jose,CA).(https://www.daowen.com)
