2.1　Cells Culture and Drug Treatment

Rattus pancreaticβ-cell line，Rin-m5F，was obtained from tumor cells bank of Chinese Academy of Medical Sciences(CAMS).Rin-m5F cells were cultured in RPMI1640 medium with 12% fetal calf serum，which adjusted to contain 2mM L-glutamine，1.5g/L sodium bicarbonate，10mM glucose，10mM N-(2hydroxyethyl)piperazine-N＇-2-ethanesulfonic acid(HEPES)，and 1.0mM sodium pyruvate.Cells were subcultured every 3～4d，two to three times weekly.Rin-m5F cells(after passages 4)were seeded into 6-well plate at a density of 1×106 per well and grown for 24h，then divided into 9 groups：control group(contained 10mM glucose)，high glucose group(contained 25mM glucose)，cytokines group(contained 10mM glucose，50U/mL IL-1β，1000U/mL TNF-α，1000U/mL IFN-γ)，high glucose+ginsenoside Rg1 group(contained 25mM glucose and 20mM Rg1)，high glucose+ginsenoside Rb1 group(contained 25mM glucose and 20m M Rb1)，high glucose+Rg1+Rb1 group(contained 25mM glucose，20mM Rg1 and 20mM Rb1)，cytokines+Rg1 group(contained cytokines motioned before and 20m M Rg1)，cytokines+Rb1 group(contained cytokines and 20m M Rb1)，cytokines+Rg1+Rb1 group(contained cytokines，20mM Rb1 and 20mM Rb1)，and cultured for 48h.(https://www.daowen.com)
