Preparation of cellulase for separation and purification
The spore suspension was inoculated on 1 litre glycerol medium in a 5 litre flask，and grown at 28℃for 48 h.The mycelium was washed with 2 volumes phosphate buffer，transferred to the induction medium，and cultured with sophorose（5×10-5 M）for another 24 h.The culture filtrate was precipitated with ammonium sulphate（80%saturation），desalted through Sephadex G-25，and lyophilized.The cellulase powder was then dissolved in acetate-NaOH buffer（0.1 M，pH=5.0）for separation.(https://www.daowen.com)
