Induction

The washed mycelium was resuspended in 50 ml 0.01 M[2]phosphate buffer（pH=5.0）.Then，2.5 ml of the mycelium suspension（—10 mg dry weight）was transferred to 2.5 ml induction medium（glycerol medium minus glycerol and peptone）and 0.1 ml of sophorose（sophorose was isolated and purified from pods of Sophora japonica L，［5］final concentration：5×10-5—5×10-4 M）in a 25 ml culture flask，and incubated in a reciprocating shaker（108 strokes per min）at 36℃.The culture filtrate（0.1 ml）was harvested at different times of incubation for the cellulase assay.Under these conditions mycelium could not grow but could synthesize cellulase rapidly following induction by sophorose.The incubation period usually did not exceed 24 h.(https://www.daowen.com)
