3.8 Evolutionary Relationships with Intergenic Spacers

There are 15 intergenic(or overlapped)spacers in the 8 analyzed Hepialoidea mt genomes.Nine are longer than 10bp:trnI—trnQ,trnC—trnY,trnY—cox1,trnA—trnR,trnR—trn N,trnE—trn F,trnF—nad5,trn H—nad4 and trnS2—nad1.However,these long spacers are mostly distributed in E.signifier and N.hunanensis.There is only one spacer longer than 10bp,and it is located between trnS2 and nad1 in the host insect of O.sinensis,in T.gonggaensis and T.renzhiensis.Compared with the overall intergenic patterns among the Hepialoidea, this moth was most similar to T.gonggaensis and T.renzhiensis,with<2bp of difference distributed in 3 and 4 loci,respectively(Fig.8).The greatest difference was observed from N.hunanensis and E.signifier, with 9 and 7 loci containing≥5bp difference compared to the host insect of O.sinensis,respectively(Fig.8).The most variable locus of N.hunanensis was observed to differ from the host insect of O.sinensis by up to 42bp in length(Fig.8).These findings further support the conclusion that this host insect of O.sinensis evolves as Thitarodes, while E.signifer and N.hunanensis form as an independent lineage.
