(三)评价

Sost敲除纯合,杂合小鼠及野生型对照小鼠经PCR反应鉴定基因型。Northern blot分析Sost敲除小鼠缺乏Sost mRNA的表达。在4.5~6.5月龄的小鼠中,Sost敲除小鼠与野生型小鼠在表型上未发现明显区别,但在10月龄的基因敲除小鼠,表现出并指、面瘫及听力损害。在雄性基因敲除小鼠中,腰椎及腿部骨密度增加,μCT显示:股骨远端的干骺端区骨小梁数目增加。无论雄性或雌性转基因小鼠的皮质骨厚度均较野生对照组增加。Sost敲除转基因小鼠具有骨硬化性骨化病的高骨量表型,骨硬化性骨化病是人类罕见的遗传性疾病,Sost基因敲除小鼠的应用将有助于研究这一疾病中的分子和细胞方面的机制。
