5.2 DNA isolation，genome sequencing and assembly
Genomic DNA of G.lucidum was isolated by improved cetyl trimethylammonium bromide(CTAB)method［40］and sequenced using a whole-genome shotgun strategy.All data were generated by paired-end sequencing of cloned inserts with two different insert sizes(200bp，6000bp)using Illumina Hiseq2000 Sequencer at BGI-Shenzhen.After removing the low complexity，low quality，adapter and duplication contamination raw reads，the clean reads were assembled using the whole-genome do novo assembler SOAPdenovo［41］.(https://www.daowen.com)
