A glass column packed with 100.0mg of cordycepin molecularly imprinted polymer was used as solid phase extraction(SPE)column.Pretreatment was performed with methanol.Two kinds of samples solution were tested.The performance of the cordycepin molecularly imprinted polymer solid phase extraction system was investigated by the addition of 100μg/mL of cordycepin and adenosine to determine the adsorption rate and recovery and an extraction solution of Cordyceps militaris was used to evaluate its use for practical application.
The 1.0mL of the mixture of cordycepin and adenosine was loaded on the column at a flow rate of 0.5mL/min.The column was washed with 2.0mL distilled water，and ethanol/water(75∶25)was used to elute cordycepin.The concentrations of cordycepin and adenosine in the eluent were determined by HPLC.(https://www.daowen.com)
The extraction solution of Cordyceps militaris was evaporated to dryness and dissolved in ethanol.The 1.0mL of this solution was loaded on the molecularly imprinted polymer column at a flow rate of 0.5mL/min.The permeate from the column was collected and analyzed by HPLC.Each measurement was carried out in triplicate.
