1.3.1 斑马鱼模型
Flk1:绿色荧光蛋白斑马鱼(Flk1:GFP),内皮细胞表达GFP(由清华大学发育生物学实验室提供)。通过显微注射,将10-二十八烷基-3,3,30,30-四甲基-吲哚羰花青素-LDL(DiI-LDL,200 mg·ml-1,美国Biome dical Technologies Inc.)注射到受精后(h.p.f.)后48 h Flk1:GFP胚胎的循环血液中。对照组注射磷酸盐缓冲液(phosphate buffer saline,PBS),注射和成像部位如图1-10所示。麻醉斑马鱼胚胎[短时间暴露于0.02% Tricaine(三卡因)],用淡水唤醒,然后放入到鱼缸中。通过长时间暴露于0.02%的Tricaine使斑马鱼安乐死。

斑马鱼的胚胎用1.5%的低熔点琼脂糖固定,以维持正常的血液循环。使用徕卡LSCM(SP5)筛选直段和分叉血管段中的LDL迁移率。绿色荧光的激发波长为488 nm,红色荧光的激发波长为561 nm。使用10倍物镜记录,每次扫描512像素×512像素的焦平面。
